Senolytics: Cleaning Senescent Cells as a Way to Slow Aging

Senolytics is a relatively new therapeutic approach based on removing senescent cells (zombie cells) from the body.
These cells are known for secreting inflammatory substances and other harmful factors that contribute to the aging process and age-related diseases.

Here are some of the potential benefits of senolytics:

  • Slowing the aging process: Removing senescent cells slowed signs of aging mainly in mouse experiments. In humans, it has so far only been shown that senolytics reduce the number of senescent cells, and this only in small, early studies. The effect on slowing aging itself in humans has not yet been proven.
  • Improving overall health: Senolytics are being studied as an approach that may support many body systems, including the immune system, nervous system, and cardiovascular system, but this is mainly at the research stage.
  • Preventing diseases (research hypothesis): There is a hypothesis, based mainly on cell and animal studies, that removing senescent cells may delay age-related diseases. To date, no clinical trial in humans has proven that senolytics prevent heart disease, Alzheimer's, or other diseases. This should not be seen as a promise for disease prevention.

What has been proven, and what hasn't: The only senolytic combination tested in humans that showed a reduction in the number of senescent cells is Dasatinib, a prescription drug, together with Quercetin - and this in small, early trials. The over-the-counter supplements marketed here (quercetin/fisetin capsules, etc.) are not identical to the prescription drug combination, and their effectiveness in slowing aging in humans has not been proven. It is also worth noting that the flagship drug in the field, UBX0101 from Unity Biotechnology, failed a phase 2 trial for treating knee osteoarthritis (2020). That is, the field is still experimental.
