The paradox: why precisely the lack of zombie cells causes wounds that do not heal in the elderly

For years we assumed that zombie cells were always bad. A new study in Aging Cell changes the story: sometimes the problem is actually the lack of them.

For the past few years, the narrative has been clear: zombie (sentient) cells are bad. Get rid of them, and the body will be young again. But a new study published in Wiley's Aging Cell presents a much more complex picture: zombie cells in the right amount and at the right time are actually essential for healing. And with the elderly, the problem is that there are too few of them, not too many.

The new study challenges

The team studied wound healing in mice, comparing young and old. Results:

  • In young mice: the wounds closed within a standard time
  • In old mice: the wounds had difficulty closing

The surprising finding: temporary sensation helps!

In the young skin, after the injury, the team detected a temporary wave of senescent cells:

  • Sensation markers (p16, p21) temporarily increased
  • Beneficial SASP secreted (tissue repair factors)
  • After the wound closed, the senescent cells disappeared

In other words: The temporary sensation was part of the solution.

What happened to the beard?

In beard skin, this response was defective: fewer senescent cells, and those that were, acted differently. More chronic inflammation, less tissue repair.

The nuance: sensation is not one

There are two types of sensation: temporary beneficial (tissue repair, vaccination, development) and chronic harmful (inflammation, damage). General senolytics kill everyone, including the beneficial ones.

Practical implications

  • Senolytics (Pistin, D+Q) Do not take after surgery or injury. Let the body heal first
  • Chronic wounds need a nuanced approach, not blanket killing
  • The new approach: "Senomorphics", drugs that change the behavior of senescent cells, do not kill

The bottom line

Science ranges from simple models ("good vs. evil") to nuanced models. Biological complexity requires a nuanced approach. This is the next step of personalized medicine.
